First attempt casing, second at casing and cloning - Mushroom Cultivation (2024)

First attempt at using WBS. B+ with 50/50 tek, microwave pasteurization tek. Shotgun style chamber.

First attempt casing, second at casing and cloning - Mushroom Cultivation (1)

Simple sterilization techniques for inoculating jars. after many failed attempts with glove box and flame sterilization, i tossed the glove box and went for a tek where i use 99% iso and cotton balls. i swab the noc port (two layers tyvek on inside, jar lid with hole stuffed with poly, micro pore tap on top of poly to hold it in place) with 99% iso and leave cotton ball there. I sterilize the the needle with a quick flame then wrap the needle in 99% iso cotton ball which overs full length of needle. when i inoculate i ensure that at no point the needle is in contact with air. First attempt i got 36/37 successful jars (one pf tek jar failed, mold seems to have come from being tipped around since the mold was growing on the top of the verm layer) Second inoculation attempt with this tek gave me 30/30 successful jars.

all my casings were watered every second day by putting about half an oz of water into the sides of the casing so it could absorb through the bottom and sides. dunking after flushes produced the best results, i guess my misting techniques leave something to be desired. The cakes i has a heavy layer of verm on top which i pooled water into, unless fruits were coming string from the top. As long as the water wasnt touching the base, they didn't seem to abort and they grew big and meaty.

I realize i let them go past edible range, so i am now adjusting to pic before spores start to drop. Total harvest from the small tins and cakes was 7.5 cracker dry ounces (three flushes, some four but the trich showed up everywhere)

i followed the easy cloning tek with four of my largest fruits. sterilized needle was pressed through the stem of a iso rubbed down stem and injected into 4 separate jars of honey and water. 3 failed and once succeeded. i inoculated 30 jars (half WBS half corn) (as described above) with the good jar, and they all succeeded. I then made 3 new jars out of the one good one, they all succeeded, and refrigerated one

with the 30 jars i made 6 casings in 10.5 by 14.5 inch foil roast pans, 2.5 inches deep. I figured i might as well cut out the small dishes as i was probably restricting the size of my fruits due to water available

i used gypsum at 10%, coffee at 10%, coco at 40% and verm at 40%. Pasteurized in a pot of water according to RR's tek, for fear of screwing up with using coffee for the first time, not sure i trusted microwave tek this time around. Casing was same mix, except without the coffee, a little more verm, and the addition of oyster shell crushed, which i ground finer in my coffee grinder to make the material more quickly useful at increasing ph. PH was around 7.5 for a half inch casing layer.

those two in the 85 liter FC are made with WBS, in a 110 liter tote i have 3 of the same size casings with corn, and pftek as spawn. they were 2 days behind the WBS and arent pinning as well, will update soon about it, not sure if the different FC is causing the problems or if its corn, or if its because the clone came off a WBS casing and has aken to that media better. bulk substrate and casing recipes identical to that of the WBS casings

I would say there is much fail here. But I don't argue with success.
You did everything wrong. And grew a bunch of shrooms.
Nice grows.

I dont understand how you could get such a terrible failure rate using a glovebox.

Unless you were trying to use one of those old iffy teks that tell you to put a fan on it with a so called cheap hepa filter.

or your sterile procedures just wernt right.

Im the opposite, in open air, contam city. SAB 99% success. Plus im confident i can open a jar if i need to and it shouldnt contam.

Open a jar in open air for a few seconds and see what happens.

Failure rates were high in my glove box probably due to one of two things, or both. my box was poorly built, not perfectly sealed from all air, and two, i was using spore syringe of GT from a vendor i wont name, but 6/6 of his GT syringes got cobweb, black mold, or trich. I assume vendor fault as the contams were all in the injection area. new syringes were sent to make up for it.

my open air with flame and 99 alcohol led to 66/67 jar success

what reason do i have to open a jar of raw grain? i wouldn't find myself doing a grain to grain, if i recall from a post from RR once, he talked about injecting water into a 100% colonized jar, shake it up, then pull a syringe from that to make as many jars as you want.

all procedures i used were from teks posted onto the shroomery
